The effect of endothelin 1 (ET-1) on the vascular smooth muscle was examined in isolated rat main pulmonary artery (MPA). The rings were precontracted by 10(-5) M phenylephrine and the constrictor response was obtained by cumulative doses of ET-1. ET-1 caused concentration-dependent increases in force of contraction in the precontracted MPA, with an IC50 value of 0.7 nM, and this increase in the force of contraction was antagonized by the specific ET-A antagonist BQ123, but not by the ET-B receptor antagonist BQ788. BQ123, as well as the ET-B receptor agonist sarafotoxin S6c (1-10 pM), also caused concentration-dependent increases in force of contraction. The MPA rings were preconstricted by 10(-5) M phenylephrine and then incubated in a Tyrode's solution containing 10(-6) M ET-1, 10(-6) M ET-1 plus 10(-7) M BQ123 or 10(-6) M ET-1 plus 10(-7) M BQ788 for 15 min. Thereafter, 10(-6) M ET-1 again caused concentration-dependent increases in force of contraction in the preconstricted MPA, with an IC50 value of 1.6 nM. The response to ET-1 was significantly reduced by incubation with BQ123 (10(-7) M) but not with BQ788 (10(-7) M).
